Special Bug Repellent

The quest can't be finished until after you finish the story but you can pick it up at any time. Speak with Tonjiro in the Elder's house who will tell you about recipe to Granny Osen's special Bug Repellent. He says he'll tell you the secret of how it's made if you tell him the secret of Karakuri Mansion. If you return to him after finishing the story he'll tell you the recipe and Bufu's Cave will unlock north of the Ruined Temple on the world map. Apparently the recipe for the special bug repellent is Daze Radish, Splitter, and Dark Komuso meat and Granny's spit. Bringing these 3 meats to Granny Onsen in the house just south of the entrance to the village will reward you with insect repellent. Never talk to Granny Onsen with only 1 or 2 of the meats in your inventory. If you do, she'll take them and they will not be used to progress the quest. Bring it to the past and hand it to the man in front of the tree. When you return to the current time, the tree will be revived and the man standing in front of it will give you a Tree Shield and the Millennium Pill. Finishing this quest is the second requirement to unlock the Tsuzura Maze.
